I would imagine so. That's how we use them, most houses are wired that way and most consumer uses will be linking upstairs to downstairs. It's not a big expense if it doesn't but if I understand what you asked and how you are wired, I'm 99% sure it'll be ok

Best thing I ever did was use a powerline to extend our wifi. Now have it all over the house and in the garden. Much better than a wireless extender.
